Knockdown of the E3 ubiquitin ligase UBR5 and its role in skeletal muscle anabolism

چکیده

UBR5 is an E3 ubiquitin ligase positively associated with anabolism, hypertrophy, and recovery from atrophy in skeletal muscle. The precise mechanisms underpinning UBR5’s role the regulation of muscle mass remain unknown. present study aimed to elucidate these by silencing gene vivo. To achieve this aim, we electroporated a UBR5-RNAi plasmid into mouse tibialis anterior investigate impact reduced on anabolic signaling MEK/ERK/p90RSK Akt/GSK3?/p70S6K/4E-BP1/rpS6 pathways. Seven days after RNAi electroporation, although reductions overall were not detected, mean cross-sectional area (CSA) green fluorescent protein (GFP)-positive fibers (?9.5%) number large lower versus control. Importantly, significantly total RNA, synthesis, ERK1/2, Akt, GSK3? activity. Although p90RSK phosphorylation increased, levels demonstrated 45% reduction UBR5-RNAi. Finally, early events 7 knockdown culminated significant (?4.6%) larger fiber CSA (?18.5%) 30 days. This was increased phosphatase PP2Ac inappropriate chronic elevation p70S6K rpS6 between days, as well corresponding eIF4e. demonstrates that plays important anabolism/hypertrophy, whereby culminates atrophy.

Modulation of myocardin function by the ubiquitin E3 ligase UBR5.

Fully differentiated mature smooth muscle cells (SMCs) are characterized by the presence of a unique repertoire of smooth muscle-specific proteins. Although previous studies have shown myocardin to be a critical transcription factor for stimulating expression of smooth muscle-specific genes, the mechanisms regulating myocardin activity are still poorly understood. We used a yeast two-hybrid scr...

Functional Roles of the E3 Ubiquitin Ligase UBR5 in Cancer.

The Ubiquitin-Proteasome System (UPS) is an important regulator of cell signaling and proteostasis, which are essential to a variety of cellular processes. The UPS is disrupted in many diseases including cancer, and targeting the UPS for cancer therapy is gaining wide interest. E3 ubiquitin ligases occupy a key position in the hierarchical UPS enzymatic cascade, largely responsible for determin...

Identification of phosphorylation sites on the E3 ubiquitin ligase UBR5/EDD.

UBR5 (ubiquitin protein ligase E3 component n-recognin 5)/EDD (E3 ligase identified by differential display) is an E3 ubiquitin ligase that is a potential biomarker for poor prognosis for recurrent, platinum-resistant ovarian cancer. UBR5 has a role in the DNA damage response and many such proteins are regulated by phosphorylation. UBR5 is a 309 kDa nuclear phosphoprotein that we previously ide...

The E3 ubiquitin ligase TRIM62 and inflammation-induced skeletal muscle atrophy

INTRODUCTION ICU-acquired weakness (ICUAW) complicates the disease course of critically ill patients. Inflammation and acute-phase response occur directly within myocytes and contribute to ICUAW. We observed that tripartite motif-containing 62 (TRIM62), an E3 ubiquitin ligase and modifier of inflammation, is increased in the skeletal muscle of ICUAW patients. We investigated the regulation and ...
